Dolonil SR100 mg
ACME Laboratories Ltd.

TRD-Contin 100 mg is a centrally acting opioid analgesic prescribed for the management of moderate to severe pain, whether acute or chronic. It is widely used when non-opioid pain relievers are insufficient to control discomfort.
Common conditions in which Tramadol is indicated include:
Tramadol may be used alone or, in certain formulations, in combination with other analgesics to enhance pain relief. It should always be taken under the supervision of a registered physician.
Opioid analgesics
Tramadol is a centrally acting synthetic analgesic with a dual mechanism of action that distinguishes it from conventional opioids. It provides pain relief through two complementary pathways working together.
First, Tramadol and its active metabolite bind to and stimulate mu-opioid receptors in the central nervous system, reducing the perception of pain. Second, it inhibits the reuptake of the neurotransmitters serotonin and noradrenaline (norepinephrine), thereby enhancing the descending inhibitory pain pathways in the spinal cord.
By activating both serotonergic and noradrenergic pathways while modulating central monoaminergic inhibitory systems, Tramadol modifies the transmission of pain impulses to the brain. This combined opioid and monoaminergic activity delivers effective analgesia with a comparatively lower risk of respiratory depression than traditional strong opioids at equivalent doses.
The dose and dosing frequency of TRD-Contin 100 mg depend on the severity of pain, the patient's response, and the dosage form used. It should be individualised by a registered physician and adjusted to the lowest effective dose. The total daily oral dose should not exceed 400 mg.
Doses should be taken with water and may be taken with or without food. Sustained-release forms must be swallowed whole and never crushed or chewed. Always follow the exact instructions of your prescribing physician.
In general, clinically significant interactions with Tramadol are uncommon, but certain combinations require caution or should be avoided:
Always inform your doctor about all prescription medicines, over-the-counter drugs, and herbal supplements you are taking before starting Tramadol.
TRD-Contin 100 mg should not be used in the following situations:
Tramadol should also be avoided or used only with specialist caution in patients with a history of seizure disorders or opioid dependence.
Like all medicines, TRD-Contin 100 mg may cause side effects, though not everyone experiences them. Most are mild and tend to lessen as the body adjusts.
Common side effects include dizziness or vertigo, nausea, vomiting, constipation, headache, drowsiness (somnolence), itching (pruritus), sweating, dry mouth, indigestion (dyspepsia), diarrhoea, weakness (asthenia), and central nervous system stimulation.
Less common side effects include general malaise, allergic reactions, weight loss, flushing (vasodilatation), palpitations, abdominal pain, loss of appetite (anorexia), flatulence, gastrointestinal bleeding, hepatitis, and inflammation of the mouth (stomatitis).
Seek immediate medical attention if you experience difficulty breathing, seizures, severe allergic reactions, or signs of serotonin syndrome such as agitation, rapid heartbeat, or high fever.
The safe use of Tramadol during pregnancy has not been established. Tramadol crosses the placenta, and there are no adequate, well-controlled studies in pregnant women. It should therefore be used during pregnancy only if the potential benefit clearly justifies the possible risk to the foetus.
Tramadol should not be used during breastfeeding, as the drug and its metabolites pass into breast milk and may affect the nursing infant. Consult your physician before using Tramadol if you are pregnant, planning a pregnancy, or breastfeeding.
TRD-Contin 100 mg should be used with care in certain patients and situations:
Avoid driving or operating machinery until you know how Tramadol affects you, as it may cause drowsiness or dizziness. Do not stop long-term treatment abruptly without medical advice.
Children: In children from the age of 1 year, TRD-Contin 100 mg may be given at a dose of 1–2 mg/kg body weight. However, the 100 mg suppository should not be used in children and adolescents below 14 years of age.
Sustained-release forms: Tramadol 100 mg SR capsules have not been studied in children. Their safety and efficacy in this group have not been established, so these products should not be used in children.
Elderly and patients with kidney or liver impairment: Dose reduction and careful monitoring are recommended, as clearance of the drug may be reduced in these patients.
Keep the medicine below 30°C, protected from light and moisture. Store in a dry place away from direct sunlight. Keep out of the reach of children and pets. Do not use after the expiry date printed on the packaging.

Opioid analgesics
Tramadol is a centrally acting synthetic analgesic compound. It inhibits the re uptake of neurotransmitters- serotonin and noradrenaline. Thus it modifies the transmission of pain impulses by activating both descending serotonergic pathways and noradrenergic pathways involved in analgesia. The analgesic effects of Tramadol are mediated via stimulation of mu-opioid receptors and indirect modulation of central monoaminergic inhibitory pathways.
Safe use of Tramadol in pregnancy has not been established. Tramadol has been shown to cross the placenta. There are no adequate and well-controlled studies in pregnant women. Therefore, Tramadol should be used during pregnancy only if the potential benefit justifies the risk to the foetus. TRD-Contin 100 mg should not be administered during breast feeding as Tramadol and its metabolites have been detected in breast milk.

TRD-Contin 100 mg 100 mg SR Capsules have not been studied in children. Therefore, safety and efficacy have not been established and the product should not be used in children.
