About Avanafil

Indications of Avanafil

Avanafil is a phosphodiesterase type 5 (PDE5) inhibitor that is FDA-approved for one established indication:

  • Erectile dysfunction (ED) in adult men — Avanafil is indicated for the treatment of erectile dysfunction, a condition in which a man cannot achieve or maintain an erection sufficient for satisfactory sexual activity. Avanafil works only in the presence of sexual stimulation and does not increase sexual desire.

Avanafil is not indicated for use in women or children, and it does not treat premature ejaculation, low libido, or infertility. It also does not protect against sexually transmitted infections (STIs), including HIV.

Dosage & Administration of Avanafil

Dosing for Erectile Dysfunction

PopulationRecommended Dose
Adult men (starting dose)100 mg taken approximately 15–30 minutes before anticipated sexual activity, as needed
Dose range50 mg to 200 mg, adjusted based on individual efficacy and tolerability
Maximum frequencyOne dose per day; not for continuous daily use
With moderate CYP3A4 inhibitors (e.g. erythromycin, fluconazole, diltiazem)Maximum 50 mg in a 24-hour period
With alpha-blockersStart at 50 mg once alpha-blocker therapy is stable (see Interactions)
Severe hepatic or renal impairmentAvanafil is not recommended (see Use in Special Populations)

Avanafil may be taken with or without food. Sexual stimulation is required for Avanafil to produce an erection. Avanafil should not be used more than once daily, and it is not indicated for use in women or in patients under 18 years of age.

Side Effects of Avanafil

The most commonly reported side effects of Avanafil (occurring in approximately 2% or more of users) include:

  • Headache
  • Flushing (facial redness/warmth)
  • Nasal congestion
  • Nasopharyngitis (common cold-like symptoms)
  • Back pain
  • Dizziness

Less common but more serious effects that require prompt medical attention include:

  • Prolonged, painful erection (priapism) lasting more than 4 hours.
  • Sudden decrease or loss of vision in one or both eyes.
  • Sudden decrease or loss of hearing, sometimes with ringing in the ears (tinnitus) or dizziness.
  • Chest pain, palpitations, or symptoms of a heart problem during or after sexual activity.

Most side effects of Avanafil are mild to moderate and transient. If any side effect is severe, persistent, or concerning, medical advice should be sought.

Contraindications of Avanafil

Avanafil is contraindicated in the following situations:

  • Known hypersensitivity to Avanafil or any component of the tablet.
  • Concurrent use of organic nitrates, in any form, whether regularly and/or intermittently — this combination can cause severe, potentially fatal, hypotension.
  • Concurrent use of guanylate cyclase (GC) stimulators such as riociguat, for the same reason (risk of severe hypotension).
  • Concurrent use of strong CYP3A4 inhibitors (e.g., ketoconazole, itraconazole, ritonavir, clarithromycin), which markedly raise Avanafil blood levels and increase the risk of adverse effects.

Frequently Asked Questions about Avanafil

What is Avanafil used for?

Avanafil is used to treat erectile dysfunction (ED) in adult men, helping them achieve and maintain an erection sufficient for sexual activity when combined with sexual stimulation.

How should I take Avanafil?

Avanafil is usually taken as a single 100 mg dose about 15–30 minutes before sexual activity, with or without food, and should not be taken more than once a day. Your doctor may adjust the dose between 50 mg and 200 mg based on your response.

Can I take Avanafil with nitrate medicines?

No. Taking Avanafil together with nitrate medicines (such as nitroglycerin, used for chest pain) or guanylate cyclase stimulators like riociguat is strictly contraindicated, as this combination can cause a severe, potentially fatal, drop in blood pressure.

What are the common side effects of Avanafil?

The most common side effects of Avanafil include headache, flushing, nasal congestion, back pain, and dizziness. Most are mild and temporary, but an erection lasting more than 4 hours, or sudden vision or hearing loss, needs immediate medical attention.

Can women or children use Avanafil?

No. Avanafil is not indicated for use in women or in individuals under 18 years of age; its safety and effectiveness have not been established in these groups.

Does Avanafil protect against sexually transmitted infections?

No. Avanafil treats erectile dysfunction only; it does not protect against sexually transmitted infections (STIs), including HIV, so appropriate precautions should still be used.
